Circular dichroism and scanning probe microscopy were used to characterize the interaction of DNA with the nonhistone chromatin protein HMGB1 and its recombinant version HMGB1(A+B) devoid of the C-terminal acidic region. The AFM data corroborate the earlier suggestion concerning the action of tandem DNA-binding domains, and support a modulatory role of the C-terminal domain in HMG protein-DNA interactions.
